Investigating methods of inducing prediabetes and diabetes in small mice

Abstract
Prediabetes and diabetes are conditions in which blood sugar levels are higher than normal. Prediabetes is a stage that occurs before diabetes and can be prevented by changing lifestyle, diet, and physical activity. Type 1 and 2 diabetes are different types of diabetes in which the body cannot produce or use insulin and insulin resistance develops, which can be dysfunctional through the high content of fatty acids and lipid-related metabolites, adipokines, and inflammatory mediators: or mitochondrial defects and endoplasmic reticulum stress. Insulin is a hormone that controls blood sugar. To study and investigate the effects and complications of pre-diabetes and diabetes, researchers use small mice such as C57 as an animal model. To induce pre-diabetes and diabetes in mice, there are various methods, some of which are: feeding with a high-fat diet, feeding with a high-sugar diet, streptozotocin injection, alloxan injection, nicotinamide injection, and cytokine injection. The effects that these methods cause in animal models include weight gain, decreased insulin sensitivity, increased blood sugar, damage to insulin-producing cells in the pancreas, and oxidative stress in pancreatic beta cells. The doses used in streptozotocin injection and alloxan injection are usually 40 to 65 mg intravenously, and in nicotinamide injection, the dose is 110 to 150 mg intravenously. In these methods, it usually takes a few weeks to a few months for the mice to show a state of pre-diabetes or diabetes. Therefore, to cause diabetes or pre-diabetes, the methods of feeding with a high-fat diet, feeding with a high-sugar diet, streptozotocin injection, alloxan injection, nicotinamide injection, and cytokine injection can be used in small mice.
