The effect of an intense resistance activity session with glutamine supplementation on the relative gene expression of alpha and IIX isoforms of fast-twitch myosin heavy chain in adult male Wistar rats

Background & Objective: A session of intense resistance training causes different changes in the level of muscle cells, but so far, its effect with glutamine supplementation on the expression of myosin alpha heavy chain genes and type IIX fast twitch motor unit is less known.
Methods: 30 8-week-old male mice with an approximate weight of 220 ± 20 were prepared and divided into three control groups, intense resistance training and intense resistance training with glutamine supplementation, in a simple random manner. The training groups participated in a resistance activity session of climbing the ramp with 4 sets, 5 repetitions, 30 seconds of rest between repetitions and 2 minutes of rest between sets. Glutamine supplement was once a day powder dissolved in 100 cc of distilled water at a dose of 5.5 grams per kilogram of body weight every day for 5 days. The extensor Digitorum Longus muscle tissue was sent to the relevant laboratory for gene expression studies of alpha and IIX isoforms of myosin heavy chain. The relative fold check method was used to check the gene expression data at a significance level of 5%.
Results: The present study showed that a session of intense resistance activity causes a significant increase in the genes of myosin alpha heavy chain (P ≥ 0.001) and type IIX fast-twitch motor unit (P ≥ 0.001). In addition, simultaneous intake of glutamine supplement also increased the values of these genes compared to the control group (P ≥ 0.016). On the other hand, the alpha isoform gene changes were significant only in the training groups alone compared to the group of glutamine combination and aerobic training (P ≥ 0.009).
Conclusion: It seems that a session of intense resistance activity is more effective than intense activity with glutamine supplementation on the increase of alpha heavy chain gene, but it has no effect on isoform IIX.
